چکیده

Drugs that target protein kinase C (PKC) are now being evaluated in patients with non-small cell lung cancer (NSCLC), but the role of PKC in NSCLC cells remains unclear. We report here that NSCLC cell lines show enhanced phosphorylation and altered expression of specific PKC isoforms compared with normal lung epithelial cells. PKC inhibition variably increased apoptosis, with rottlerin, a PKC inhibitor, being most effective and potentiating chemotherapy-induced apoptosis, especially with trastuzumab. Consistent with PKC being anti-apoptotic in NSCLC cells, transient transfection of a kinase-dead mutant of PKC increased apoptosis and potentiated chemotherapy-induced apoptosis. Our studies provide a rationale for targeting PKC isoforms in NSCLC cells, especially PKC .

Protein Kinase C and Apoptosis

The protein kinase C (PKC) family consists of ten structurally related serine/threonine protein kinases. PKC isoforms are critical regulators of cell proliferation and survival and their expression or activity is altered in some human diseases, particularly cancer. The development and utilization of PKC isoform specific tools, including dominant inhibitory kinases, mouse models in which specifi...

Distinct PKC isoforms mediate cell survival and DNA synthesis in thrombin-induced myofibroblasts.

Thrombin activates protease-activated receptor (PAR)-1 and induces a myofibroblast phenotype in normal lung fibroblasts that resembles the phenotype of scleroderma lung fibroblasts. We now demonstrate that PAR-1 expression is dramatically increased in lung tissue from scleroderma patients, where it is associated with inflammatory and fibroproliferative foci. We also observe that thrombin induce...

Carvacrol and Thymol Attenuate Cytotoxicity Induced by Amyloid β25-35 Via Activating Protein Kinase C and Inhibiting Oxidative Stress in PC12 Cells

Background: Our previous findings indicated that carvacrol and thymol alleviate cognitive impairments caused by Aβ in rodent models of Alzheimer's disease (AD). In this study, the neuroprotective effects of carvacrol and thymol against Aβ25-35-induced cytotoxicity were evaluated, and the potential mechanisms were determined. Methods: PC12 cells were pretreated with Aβ25-35 for 2 h, followed by ...
